MAGPIX System - How To Use Protocol and Batch Function

The move from multiple protein assays to multiplex assays is now totally within reach! The MAGPIX system is an affordable, compact, fluorescent-based detection system based on Luminex's proven xMAP technology. With the MAGPIX system every researcher can perform quantitative analysis of up to 50 proteins simultaneously using only 25μl of precious sample. The MAGPIX system is personal, benchtop instrument with out of the box set-up with step by step guide to interactive software and all designed for use with broad and expanding menu of assay kits. Success in using your MAGPIX system requires that you're able to setup a Novex multiplex assay protocol and batch using your xPONENT software. To learn more about Novex singleplex and multiplex assay visit www.thermofisher.com/luminex. Before you start, remember to save the probe height and plate name in the maintenance section. This will be important later on. To start, open up the Protocol tab in xPONENT and select "Create New Protocol." To create a new Protocol name you'll need to complete the following information: Type in Batch name For Manufacturer, write "Life" For Volume: type in the number "50". Note: Volume can be adjusted for optimal performance. For Plate Name: choose the saved plate from the drop down menu. For Analysis Type: select "Quantitative" For number of Standards: type in the number 7. Note: for more standard points, type in the corresponding number Once you've filled in each field, click "Next." Now, select the desired bead regions and enter the descriptions on the right. For units "pg/ml" and for "Count", enter 100. Select Apply All. You are able to change the algorithm; however we recommend using 5PL Logistic Weighted. When you're finished, click "Next". You are now ready to select the replicate as either vertical or horizontal positioning and select the wells used for background and standards. Don't worry about any of the "unknown" wells at this time—they don't have to be completed at this step. You can choose to include a maintenance routine if you'd like, but it's not required. Remember to "Save" your protocol. Find the protocol screen and highlight the new protocol and then select "New Standard/Control". You'll have to enter details for the Name, Lot, Expiration Date, and Manufacturer fields. Once you complete this information, find the "Standard Information" field and enter the name, lot, expiration date, and manufacturer under "Standard 1". You'll want to highlight all four fields and use the down arrow to populate all the standards. For each target, you'll need to enter the highest concentration on the "standard 1" line. Highlight all the standard 1 values and then type "3" in the Apply Dilution box for Novex singleplex and multiplex assay. Select "Apply Dilution" to automatically calculate the dilutions. Next, find the "Batches" screen and select "Create new batch from existing protocol". You'll be asked to complete the new batch name and select the appropriate protocol from the list provided. The standards associated with each protocol will appear below. Click "Next". The lot number and associated information will appear in the next screen. You can choose other lots by selecting the "Apply Standard/Control Kit" button and selecting from the list. Once you've chosen your lots, click "Next". It's now time to complete the information for your "unknown" wells. Be sure to verify replicate count and then populate the plate with the appropriate unknowns. On the right hand side of the screen, complete the descriptions and include a "2" for dilution factor. You have the option to right click on the first entered dilution and Apply dilution down. Individual samples that have different dilution factors can be entered as necessary. You can Save or Run the batch directly from this screen.
