Do you need to preserve sequence accuracy? If not, a standard DNA polymerase can be used to confirm presence or absence of target DNA. If, however, you will clone or perform sequencing, choose a high-fidelity proofreading polymerase for accurate sequence. Are you working with long DNA templates or looking to cut down cycling time? If so, choose a polymerase with a high processivity and extension rate. This will ensure efficient DNA synthesis and shorter reaction times. Are you encountering non-specific amplification? If you’re seeing too many bands that should not be there, choose a hot-start polymerase. This can help get more yield and sensitivity of the target.
