For a demonstration of the MAGPIX System, click here! One of the most important steps in running a successful multiplex assay on your MAGPIX system is making sure you prepare your sample correctly. Just like any other experiment, you want to make sure you have cleared a section of your lab bench to work on and make sure you have everything you need on hand. Here's the list of all the items necessary for your multiplex assay. Make sure your MAGPIX System is routinely maintained, calibrated, and verified the day of use. It's also important that you make sure the samples that you'll be using are thawed immediately before your assay is set up. Remember that samples begin to degrade upon thawing, so the least amount of time your samples are at room temperature, the better. You'll need to clarify your samples by centrifugation at 14K for 10minutes. Samples can be stored on ice until they're loaded onto a plate. It's important to note that if you're using samples other than serum or plasma, you have 5mL of that media type available for standards that you'll use in an assay. Remove your Novex multiplex assay kit from the refrigerator at least one hour before you begin to allow equilibration to room temperature. While you're waiting, you can prepare the 1X wash solution by diluting 15mL of the 20X concentrate into 285mL of de-ionized water. In some cases, crystals may be present in the wash concentrate. If this occurs, warm the concentrate in a 37°C water bath until dissolved, or dilute the wash solution with crystals in 285mL of water and allow the wash solution to dissolve at room temperature. You'll want to prepare a wash station near a sink or dispensing reservoir for later use with your assay. It's also helpful to gather a small stack of paper towels that will be used for blotting your assay plate later on. You can now begin to set up the primary stage of your assay by following the steps located in the multiplex assay protocol supplied with the kit.
